Anna-Sophie Brüning finished her studies of violin and piano in Hannover, Freiburg and Chicago.
Her teachers included Rainer Kussmaul and Shmuel Ashkenasi (violin) as well as Karl-Heinz Kämmerling (piano). Awarded with several prizes, Brüning was appointed concertmaster of the Philharmonic Orchestra of Lübeck in 1999. Parallel to her engagement, she studied conducting at Musikhochschule Leipzig and graduated with honors in 2003, also winning the Leipzig Hochschulwettbewerb in conducting. Same year, she received a scholarship to join the Dirigentenforum des Deutschen Musikrats for 5 years.
During her studies she was engaged as répétiteur at Theater Lübeck and in the same position in the season 2003/04 at Staatsoper Unter den Linden Berlin. In 2005, she debuted at Komische Oper Berlin and was consecutively engaged as conductor at the same institution in seasons 2006 to 2008. She also worked with orchestras like Nürnberg Philharmonic, Stuttgart Kammerorchester, Polish Kammerorchester, Nordwestdeutsche Philharmonie, Sinfonie-Orchester Mainz, Philharmonie Gera and Lübeck Philharmonic.
In 2003, Brüning was engaged by Daniel Barenboim to help establish the first Palestinian Symphony Orchestra as an artistic director. She lived and worked in the Middle East for several years and regularly appeared with Daniel Barenboim in Palestine, Israel and Jordan.
In the season of 2007/08, she guested at Tonhalle Düsseldorf to high acclaim and was immediately reinvited to conduct Holst’s Planets in the subsequent season. Additionally, she worked with the LJO Baden-Württemberg and conducted the Christmas concert of Federal President Horst Köhler, which was broadcasted by ZDF on Christmas Eve.
Since 2007, Brüning regularly works in a close artistic relationship with prizewinning author and stage director Paula Fünfeck. Several pieces, such as Die arabische Prinzessin oder das wiedergefundene Leben (The Arabic Princess or the recovered life), have been written, arranged and published by Boosey&Hawkes and been brought to stages in Leipzig, Bonn, Graz, and Bilbao.
Brüning gave her debut with La Traviata at Nationaltheater Mannheim in 2009, and also conducted several concerts with the Brandenburg Symphonic and the orchestra of Landestheater Flensburg. In 2010, she conducted at Landestheater Oldenburg (Troubadour/Verdi), at Theater Pforzheim (Margarete/Gounod), and with the Niedersächsische Staatsorchester Hannover (Hansel and Gretel).
From 2012 to 2015, Brüning held the position of 1st Conductor at Landestheater Coburg. There, she conducted operas like Rinaldo, Don Giovanni, The Gypsy Baron, The Barber of Baghdad, Madama Butterfly, The Arabic Princess, L'elisir d'amore, Hansel and Gretel, Orfeo(Gluck), Savitri(Holst), Die Entführung aus dem Serail, as well as concerts for families and children, and symphonic  concerts with the soloist Sabine Meyer.
In the past season, among other things, she led the world premiere of the revised version of F. Schubert’s opera Die Bürgschaft, a co- production by the LJO Baden-Württemberg and the JVA Adelsheim, financed by the culture ministry of Baden- Württemberg.
She also led the project 'Der Zauberlehrling', a cooperation with the music schools of Karlsruhe and Freiburg, at the Baden-Baden Festspielhaus. The project consisted of musical workshops for children and a subsequent concert moderated by Anke Engelke.
In spring of 2016, Brüning debuted in Innsbruck with the baroque opera pasticcio Himmelsgeigen und Höllenfeuer by Heinrich Ignaz Biber, for which she herself has written the concept and the musical arrangement.
